He joined Villarreal from River Plate in January 2008 and played the next 18 months on loan with Recreativo Huelva before they were relegated in 2009.
Ruben is due to travel to England to seal a deal with the Premier League club, Villarreal said on their website.
He has also played for Rosario Central in Argentina, and has scored nine goals Villarreal B so far in the 2009-10 campaign.
Wigan, currently just three points above the relegation zone, are also reportedly one of several clubs interested in signing Rangers striker Kris Boyd.
